The real source
The claim comes from a 2025 review by Hwang and Oh, published in the International Journal of Molecular Sciences, titled "Interaction of the Vagus Nerve and Serotonin in the Gut-Brain Axis." A review does not report a brand-new discovery, it synthesizes what is already established across many studies into a clearer picture, in this case of how gut serotonin reaches the brain.
Where gut serotonin comes from
About 90 percent of the body's serotonin is made outside the brain, mostly by enterochromaffin cells lining the gut wall. These cells release serotonin in response to mechanical stretch, nutrients, and signals from gut microbes. That serotonin does not simply diffuse into the bloodstream and drift to the brain, serotonin cannot cross the blood-brain barrier in meaningful amounts. Instead it acts locally, activating 5-HT3 receptors on vagal afferent fibers, the sensory branch of the vagus nerve that runs from the gut to the brainstem.
The actual signaling path
Those vagal afferent signals arrive first at the nucleus of the solitary tract (NTS) in the brainstem, which relays the signal onward to brain regions including the dorsal raphe nucleus and the locus coeruleus, hubs involved in mood regulation, stress response, and arousal, and onward still to areas like the amygdala. Gut hormones including ghrelin, cholecystokinin and GLP-1 also modulate this vagal signaling, adding another layer to how digestion state influences brain state moment to moment.
The microbiome adds another layer
Gut bacteria influence this system too. Certain bacteria produce short-chain fatty acids that increase the activity of tryptophan hydroxylase 1, the rate-limiting enzyme the gut uses to make serotonin in the first place, meaning the composition of gut bacteria can shift how much serotonin signal is available to send.
Why this matters for neurodivergent people specifically
Gastrointestinal symptoms are reported at notably higher rates among autistic people and people with ADHD than in the general population, and mood and anxiety conditions frequently co-occur with both. This gut-vagus-brain pathway offers one plausible, evidence-backed piece of biology behind why gut symptoms and mood symptoms so often travel together in the same person, rather than being two unrelated coincidences.
What this does not mean
This is textbook-level, well-established physiology being clearly explained, not a new treatment or a promise that fixing gut health will resolve neurodivergent traits or mood conditions on its own. The pathway is real, turning that into a specific, individual intervention is a separate and much less settled question.
